Transcriptional and post-transcriptional regulation of ATP-binding cassette (ABC) transporters

Synopsis:

The study of expression and function of ABC transporters in cancer is important for two reasons: (i) ABC transporters are often expressed in tumors and other malignancies treated with anticancer drugs and easily render these tumor cells resistant to treatment within a short period of time; and (ii) the pharmacokinetics of anticancer drugs are often affected by the presence of ABC transporters in the liver and intestines, resulting in reduced oral bioavailability and increased elimination. Therefore, insights into the mechanisms governing their expression and opportunities to modulate these mechanisms are highly relevant. The overall objective of this work was thus to examine aspects of the regulation of gene expression of the ABC transporters. Given the complexity of the gene regulation processes, this work was focused on understanding the aspect of transcriptional and post-transcriptional regulation of two important members of the ABC transporter superfamily, namely BCRP and MRP1. The rationale of this work was to improve our understanding of the network of regulatory mechanisms of ABC transporters involved in drug resistance so that we can have an impact on cancer therapy.
